Armodafinil

Mechanism of Action

Armodafinil shares the same mechanism as modafinil — primarily dopamine reuptake inhibition via DAT binding — but as the isolated R-enantiomer, it has a longer effective half-life and more sustained plasma levels. It maintains wakefulness-promoting effects through histaminergic and orexinergic pathways without the shorter-acting S-enantiomer.

Dosing Research

Standard dose: 150 mg once daily in the morning (roughly equivalent to 200 mg modafinil). Available as 50, 150, 200, and 250 mg tablets. Half-life approximately 15 hours. Peak plasma concentration reached in 2 hours. Lower doses (50-150 mg) often sufficient for cognitive enhancement.

Side Effects & Risks

Similar profile to modafinil: headache, nausea, dizziness, insomnia. May cause more sustained insomnia than modafinil due to longer half-life. Same rare SJS risk. Appetite suppression common. Can interact with CYP3A4 substrates including hormonal contraceptives.
