Resveratrol

Resveratrol is a polyphenol found in red wine, grapes, and berries. Mechanism of Action
Resveratrol activates SIRT1 (directly or indirectly via AMPK), which deacetylates PGC-1alpha to enhance mitochondrial biogenesis and fatty acid oxidation. It also inhibits NF-kB to reduce inflammation, acts as an antioxidant, improves endothelial function via eNOS activation, and may mimic caloric restriction signaling. Bioavailability is poor due to rapid glucuronidation and sulfation.
Dosing Research
Common dose: 250-1000 mg/day of trans-resveratrol. David Sinclair protocol: 1000 mg/day taken with a fat source (yogurt) to enhance absorption. Micronized formulations may improve bioavailability. Often combined with NMN or NR. Some protocols use up to 2000 mg/day.
Side Effects & Risks
Generally well-tolerated. GI discomfort at high doses (>2.5 g/day). May interact with blood thinners (mild antiplatelet effect). Estrogenic activity at high concentrations (theoretical concern). Very poor bioavailability limits both effects and side effects. Quality varies significantly between supplement brands.
